Fermentation Optimization for Production of Polyhydroxybutyrate (phb) by Newly Isolated Azotobacter Vinelandii Kdp

چکیده
Producing the biodegradable plastic is the most important processes nowadays because to overcome the recent issues concerning the global environment and solid waste management. The optimum conditions for PHB production are more important to enhance the production by microorganism. In the present investigation newly isolated Azotobacter vinelandii KDP culture conditions were optimized to produce PHB. The optimum pH and temperature of media for biomass and PHB production by A. vinelandii KDP was 7.0 and 35C respectively. Maximum biomass (2.67g/l) and PBH (2.18g/l) production obtained at 5g/l sucrose contained media. 2.0 mM/l NH4Cl was the best concentration for maximum yield of PHB (1.28g/l). Optimum concentration of trace metal selenium for maximum yield of biomass and PHB production was 80μg/l. Under this optimum condition A. vinelandii produced two fold PHB.

Production of natural fiber reinforced thermoplastic composites through the use of polyhydroxybutyrate-rich biomass.
Previous research has demonstrated that production of natural fiber reinforced thermoplastic composites (NFRTCs) utilizing bacterially-derived pure polyhydroxybutyrate (PHB) does not yield a product that is cost competitive with synthetic plastic-based NFRTCs. Moreover, the commercial production of pure PHB is not without environmental impacts. To address these issues, we integrated unpurified ...

We previously showed that in Azotobacter vinelandii, accumulation of polyhydroxybutyrate (PHB) occurs mainly during the stationary phase, and that a mutation in phbR, encoding a transcriptional regulator of the AraC family, reduces PHB accumulation. In this study, we characterized the roles of PhbR and RpoS, a central regulator during stationary phase in bacteria, in the regulation of expressio...
متن کاملProduction of polyhydroxybutyrate and alginate from glycerol by Azotobacter vinelandii under nitrogen-free conditions.
Glycerol is an interesting feedstock for biomaterials such as biofuels and bioplastics because of its abundance as a by-product during biodiesel production. Here we demonstrate glycerol metabolism in the nitrogen-fixing species Azotobacter vinelandii through metabolomics and nitrogen-free bacterial production of biopolymers, such as poly-d-3-hydroxybutyrate (PHB) and alginate, from glycerol. Gl...

Several aspects of alginate and PHB synthesis in Azotobacter vinelandii at a molecular level have been elucidated in articles published during the last ten years. It is now clear that alginate and PHB synthesis are under a very complex genetic control. Genetic modification of A. vinelandii has produced a number of very interesting mutants which have particular traits for alginate production. On...

Modeling and simulation of Polyhydroxybutyrate (PHB) production by Protomonas extorquens in fed-batch culture were conducted in this research. The fed-batch model, developed for this process, employed a kinetic model proposed by other researchers. Several kinetic models were investigated to choose the best model. The criterion for this selection was goodness of fit (δ2). Haldane kinetic model w...
